Formalin Instructions for Use
Thermo Scientific Richard-Allan Scientific Concentrated Neutral Buffered Formalin is a clear colorless solution.
When diluted with 4 gallons of distilled or deionized water (ratio 1:4), the resulting composition contains 10%
formalin volume by volume. It is recommended to use distilled or deionized water instead of tap water due
to variations in pH and minerals often found in tap water. Dibasic sodium phosphate and monobasic sodium
phosphate buffers are used to assure a consistent pH of 6.8-7.2 at 25°C. The adequate buffering capacity is
maintained in each gallon of the Concentrated Neutral Buffered Formalin. The product is free from particulate
at room temperature (25°C) and stable to freezing temperatures. During winter months, however, a white
precipitate may form upon shipping. Storage at room temperature for 24 to 48 hour period will ensure elimina-
tion of the precipitate. Unlike 37% formaldehyde and some other formalin concentrates, the precipitate will go
back into solution.
Concentrated Neutral Buffered Formalin should never be used undiluted. This will result in incomplete fixation
especially within the core of the tissue. Excessive cross-linking will occur on the periphery of the tissue causing
the inner areas to be absent of fixation. This will also deter the influx of other reagents into the inner tissue
because of the excessive crosslinking matrix.
Concentrated Neutral Buffered Formalin is a good alternative to making 10% Neutral Buffered Formalin in the
lab. The time spent weighing chemicals and mixing, as well as keeping necessary raw material inventory in the
laboratory should be considered. Exposure limits are significantly reduced due to less contact with formalde-
hyde and storage space is reduced.
Instructions For Use
1. Remove cap from a suitable 5 gallon container or carboy.
2. Make sure air vent is closed.
3. Pour 1 gallon of Concentrated Neutral Buffered Formalin into container or carboy.
4. Fill container or carboy with 4 gallons distilled or deionized water.
5. Replace cap on container or carboy.
6. If the container or carboy has a spigot, position container so that the spigot overhangs on the counter.
7. Dispense liquid.
