Introduction
General
This document describes the configuration of the Varian
ProStar 325 UV-Vis Detector driver. It also explains how to
build a control method for this driver, how to start and view a
running acquisition, and what actions are available during an
analysis.

Installation and Configuration
Installing the Driver Software
To install the Galaxie Drivers software refer to the Galaxie
installation Guide.
Varian ProStar 325 Communications Configuration
Ethernet Communications
The Varian ProStar 325 UV-Vis Detector uses Ethernet as the
method of communications for instrument control.
The Varian ProStar 325 UV-Vis Detector uses TCP/IP
(Transmission Control Protocol/Internet Protocol) to
communicate over the Ethernet network and needs to be given
a unique IP address to identify the instrument. A static IP
address can be also configured in the Varian ProStar 325 with
the handheld unit.

A BOOTP Server is used to send IP addresses to TCP/IP
devices.
The BOOTP Server lists Ethernet addresses along with the IP
addresses that are to be assigned to the corresponding
devices.
To known how to configure the BOOTP server, refer to the
Galaxie installation Guide.
NOTE: The BOOTP server provided by the Galaxie Chromatography Data System is
installed during the installation of the Galaxie software.
Configuring the Driver
The driver software configuration is done in the Galaxie
Configuration Manager when a new chromatographic system is
created.
Configuring the Varian ProStar 325 UV-Vis Detector
Click on the button corresponding to the Varian ProStar
325 driver and the configuration screen appears.
The toolbar allows you to easily scroll through the configuration
windows:
to select the first configuration window.
to select the previous configuration window.
to select the next configuration window.
to select the final configuration window.

Communication Window
In the IP Address field, enter the IP address of the instrument.
Press the button
to search the subnet for all connected
Varian ProStar 325 instruments. The following window will be
displayed.

Once an IP Address is configured, press the button
to activate the communication with the
instrument.
Check the Synchronize with other modules box to enable this
option. When this option is enabled the Varian ProStar 325 will
be started from a contact closure which will be able to receive a
fault signal directly from another instrument.
Check the Enable Ready In box to enable this option. When this
option is enabled the Varian ProStar 325 waits for a ready signal
to go to the ready state.

Option Window
Press the
button to enter a code which
will enable the Dual Wavelength mode in the instrument. Then
check or uncheck the Two Wavelength Option to use it in
Galaxie.
Select the Cell Type according to the one physically installed in
the detector, four cell types are selectable: the 9x0, 9x1, 4x0
and 4x0.15.
If a prep flow cell is selected, the user will be able to enter a
Cell Ratio. The ratio is used for extended range operation. The
ratio for each flow cell is determined at the factory and is
included with the flow cell. The detector will always operate in
extended range mode with a prep flow cell.

Configuring the Synopsis
In the Overview part of the system configuration, a synopsis can
be defined for the system. This synopsis depends on the
configured devices.
1. To save the configuration of these devices, click on the OK
button.
2. Right-click on the system name in the bottom part of the
Galaxie Configuration Manager screen. A pop-up menu
appears: click on Stop. Then right-click on the system name
again, and click on Start.
3. Go back to the System Configuration screen (Properties
wizard). A graphic of the devices that comprise the system
is displayed in the synopsis screen. To configure the
synopsis as the real instrument configuration, move each
module then connect it appropriately. To do this, select one
anchor for each of the two modules (the selected anchors
become red) and click on the
button; a connection
tube will appear between the two anchors.
Click on the
to delete a selected tube; click on the
button to delete all the tubes.
Click on
to define a background color, or on to
make the background transparent.
Click on the
icon to change the grid color.
4. Once the synopsis configuration is complete, it will be
displayed in the Galaxie Chromatography Data System, in
the Systems tab. In this tab, the Overview part will show the
synopsis of the system with some parameters displayed
(temperature, flow, signal, etc.) depending on the installed
devices.

NOTE: Once the system has been created and correctly configured, it must be
associated with one or more projects in order to start some acquisitions. For
more details about the creation and configuration of a system, refer to the
Galaxie Configuration Manager User’s Guide.

Building a Method
Creating a New Method
The method contains parameters for instrument control, data
acquisition, chromatogram processing, and editing of final
results. To initially set up a method, it is not necessary to define
all of the sub-method sections; however, you must fill in the
instrument control section prior to starting an acquisition.
1. To create a new method, select the FILE / NEW METHOD
option from the Galaxie Chromatography Data System main
menu.
A wizard guide will appear and provide assistance during
the first steps of method creation:

2. Choose the system (chromatograph) associated with the
method. A method is created for a particular system. When
starting an acquisition, the name of the system that
performs the acquisition must be correct, since method
access is limited only to those methods associated with this
system.
3. Once this field is completed, click on the Next button to
move to the second step of the method creation:
4. Enter the New Method Name in the first field. In the
Description field, enter information concerning the method
(optional).
5. Once these two fields have been completed, click on the
OK button. The new method is created and is opened with
default parameters in the Galaxie Chromatography Data
System. The next step is the definition of each of the
method sections.
The instrument control section must be defined in order to
start an acquisition. In the following part of this guide, only
the instrument control section for the Varian ProStar 325
UV-Vis Detector is detailed. Other parameters such as
Injection Volume, Vial and Rack number may be necessary
and are defined in the acquisition section.

Once the new method is created, select the control section in
the lower part of the browser.

Press the button to display the System Control Method
Advanced Tools window:
to send the current method to the instrument.
to get the active method from the instrument.
to print the current method.
Click on the
button to display the Varian ProStar 325
control method sections. It contains four sub-sections: Signal,
Peak Sensor, Relay and Miscellaneous. Click on the required
sub-section to define the corresponding parameters that will be
used during the chromatographic run. The toolbar allows you to
easily scroll through the control method windows:
to select the first window of the control method.
to move back to the previous window of the control
method.
to move forward to the next window of the control method.
to select the final window of the control method.

Signal Control
Select from the list the Bunch Rate of the detector. With
increased bunch rates, less disk space is required to store the
data.
With decreased bunch rates, more data is collected. Bunch rate
should be set appropriately for the narrowest peak of interest in
the chromatogram.
Enter the Noise Monitor Length in the corresponding field. This
parameter sets the number of points collected during the
monitoring period and is used to calculate the baseline noise.
Enter the Response Time in the corresponding field. This
parameter specifies the degree of electronic noise filtering

applied to the analog outputs. It affects only the recorder and
data system analog outputs from the detector module, and does
not affect the data sent to the Galaxie Chromatography Data
System.
Enter the time program for the Wavelength, the Attenuation, and
the Autozero parameters.
The attenuation parameter switches the detector's output
attenuation to one of the eleven available settings. Attenuation is
measured in Absorbance Units Full Scale (AUFS). This affects
only the recorder’s output.
If Yes is selected in the Autozero field, the detector’s output is
automatically zeroed. This affects both the chromatogram display
and the data acquired for permanent storage.
Each step of the program includes time, wavelength, attenuation,
and autozero values.
Use the following buttons to edit the table:
to add a line to the table.
to delete a line in the table.
to clear the entire table.
If Dual Wavelength Mode is checked extra columns are added in
the table which will allow the user to enter another Wavelength,
Attenuation and an Autozero value.

Peak Sensor Control
Enter the time program for the Peak Sensor parameters.
In the Peak Relay column you can activate the Time Slice
function. Time slicing allows the Peaksense relay to be actuated
at regularly spaced intervals during the run. The time slicing can
be turned on or off (field at specified times, for example, On Time
2.5 min, Off Time 12.7 min. This parameter cannot be set at
initial conditions (time 0.00).
The Period parameter is the time between Peaksense relay
actuations during a time slice event. A relay period of 30 causes
the Peaksense relay to actuate every 30 seconds while time
slicing is turned on.

In the Peak Relay column you can also activate the Pulse Relay
function The Pulse relay activates or deactivates the Peaksense
relay at the specified time. This parameter cannot be set at initial
conditions (time 0.00).

Relay Control
This detector has four external event relays which may be time
programmed. Each step of the program includes time and state
for each relay’s new values.
Use the following buttons to edit the table:

Miscellaneous Control
Analog Control Group:
In the Analog X Source scrolling list, select Wavelength 1,
Wavelength 2, or none. These entries specify which signal will be
chosen for the two analog outputs on the rear panel of the
detector. Wavelength 2 is only useful if the detector is operating
in dual channel mode. The same signal can be output from the
two analog channels even if the detector is operating in single
wavelength mode.
In the Analog X Peak Ticks scrolling list, select enable or disable.
Peak Ticks are small spikes added to the analog output to
indicate when the Peak relay is activated.
