Preparing for Synthesis
1. Prepare an MPS Notebook and send it to
the system (see other side).
2. Print an MPS Calculation Sheet (MPS
User’s Guide).
3. Pack the columns (MPS User’s Guide).
4. Check and update the reagent resources
(MPS User’s Guide).
5. Install the amino acids in the Pioneer trans-
port system according to the Rack and Amino Acid Position diagram below (MPS User’s
Guide).
6. Prime the system and the MPS Accessory (MPS User’s Guide).
7. Install the columns on the MPS Accessory according to the Column Positions diagram
(MPS User’s Guide).
8. Empty the waste (MPS User’s Guide).
9. Start the synthesis (MPS User’s Guide).

Pioneer MPS Accessory
Quick Reference Card
Using the MPS Notebook
1. Create or open an MPS Notebook in the Database window (MPS Users Guide).
2. Enter the synthesis parameters (MPS Users Guide):
Select an MPS protocol.
Select the Chemistry.
Select the Scale.
Select the Cycle.
Select the Activator.
Select the Final Cycle option.
3. Enter the sequence for each column (MPS Users Guide, Section 2.2.3):
Click the MPS column (a through p).
Click the Sequence View button .
Type the sequence, starting with the N-terminus.
Type the sequence name in the Name text box.
Click the MPS Notebook View button .
Repeat the steps above for each MPS column (a through p).
4. Select the C-Terminus option (Acid or Amide) for each sequence.
5. Select the First AA option (On Support or Off Support) for each sequence.
6. Send the MPS Notebook to the system by clicking .
7. Prepare the System for synthesis (see other side).
