Lack of flow of all reagents to any column may indicate that the glass capillary
restrictor has become clogged. Although this part is easily replaceable, it may
often be unclogged. Two methods have been successful:
1. Sonicate in a warm water bath.
2. Boil in water.
Should these methods fail, a cleaning wire (Hamilton Company, Part Number
18300, Diameter 0.00350 inches) may be used to physically remove the clog. This
method is least desirable because it may alter the bore of the glass.
If these procedures are unsuccessful, the glass restrictors should be replaced. On
three-column instruments all three restrictors should be replaced at the same
time. Restrictors are supplied in sets of three which have been tested together
and determined to give the same flow rate. This ensures equal flow to all three
columns. Always verify the flow rate of each column.
This is done by choosing the " Control and Test Solenoid Valves" softkey on the
Model 380A or the "Manual Control" key on the Model 380B and entering Function
9 to deliver acetonitrile to a previously specified column. Remove the bottom Luer
fitting from the column and after 20 seconds of flow, collect the acetonitrile in a
graduated tube for exactly 2 minutes. The flow should be to 1.70 mL/min. ± 0.15
mL. On three column instruments measure the flow rates of all three columns
separately. If the three columns give similar flow rates but are not within the
correct range, the regulators need to be adjusted. If the three columns give
different flow rates, the restrictors need to be replaced. Call your local Applied
