GeneScan™ DS‑33 Installation Standards with GeneScan™ 500

Product description
GeneScan™ DS-33 Installation Standards with GeneScan™ 500 LIZ™ Size Standard are used to perform the install check. The GeneScan™
DS-33 Installation Standard consists of pooled PCR products that are labeled with 6-FAM™, VIC™, NED™, and PET™ dyes. To generate the
pooled products, CEPH individual 1347-02 genomic DNA has been amplied with 8 uorescently labeled PCR primer pairs that amplify
selected microsatellite loci.

Contents and storage

ready to use. Upon thawing, store the
size standard tube at 2°C to 8°C.

Prepare the installation reagent:

Genetic Analyzer
1. Resuspend the contents of the installation standard and the
size standard tubes, then centrifuge briey to bring the
contents to the boom of the tubes.
2. Combine the following components in a microcentrifuge
tube:

[1] Scale the volumes for 4-capillary instruments.
IMPORTANT! Use the installation reagent within 16 hours of
preparation.
3. Mix thoroughly, then centrifuge to bring the mixture to the
boom of the tube and eliminate air bubbles.
4. To denature the DNA fragments, incubate at 95°C for
5 minutes. Immediately place the mixture on ice for
³ 2 minutes.
5. Transfer installation reagent to the appropriate wells a 96-
well plate.
Option Description
16-capillary
instruments
Dispense 10 µL of standard into wells
A1 to H2.
4-capillary instruments Dispense 10 µL of standard into wells
A1 to D1.
6. Cover the plate with a 96-well septa (Cat. No. 4315933), then
centrifuge to bring the mixture to the boom of the tube and
eliminate air bubbles.
7. Assemble the plate with the Retainer and Base, then load on
the instrument.
8. See the instrument user guide for specics on seing up the
run.
Prepare the installation reagent: 3730xl DNA
Analyzer
Note: This installation kit is not for use on 96-capillary
instruments.
1. Resuspend the contents of the installation standard and the
size standard tubes, then centrifuge briey to bring the
contents to the boom of the tubes.
2. Combine the following components in a microcentrifuge
tube:

Hi‑Di™ Formamide 460 µL
Total 510 µL
IMPORTANT! Use the installation reagent within 16 hours of
preparation.
3. Mix thoroughly, then centrifuge to bring the mixture to the
boom of the tube and eliminate air bubbles.
4. To denature the DNA fragments, incubate at 95°C for
5 minutes. Immediately place the mixture on ice for
³ 2 minutes.
5. Transfer installation reagent to the appropriate wells a 96- or
384-well plate.
Option Description
96-well plate Dispense 10 µL of standard into every other
column (A1-H1, A3-H3,...).
384‑well plate Dispense 5 µL of standard into the
corresponding wells for a single 48-capillary
injection (A1, C1, E1, G1, I1, K1, M1, O1, A5,
C5,....).
6. Cover the plate with a 96-well septa (Cat. No. 4315933), then
centrifuge to bring the mixture to the boom of the tube and
eliminate air bubbles.
7. Assemble the plate with the Retainer and Base, then load on
the instrument.
8. See the instrument user guide for specics on seing up the
run.
Run the install standard with the Gene Mapper Software Run
Module and select Dye Set G5.
