4. Add sample type-specific buer, samples, standards, and blanks, and then incubate.
a. For serum, plasma, and CSF: Add 6.3μL of Universal Assay Buer (1X) to each well
followed by 6.3μL of prepared standards or samples into dedicated wells. For cell culture
supernatants: Add 12.5μL prepared standards or samples into dedicated wells.
b. For wells designated as blanks: Add an additional 6.3μL of Universal Assay Buer (1X) for
serum, plasma, or CSF samples. For cell culture supernatant samples, add 12.5μL of cell
culture medium.
c. Seal the plate with the provided Plate Seal. Cover the plate with the Black Microplate Lid and
shake at 1,400 ± 50 rpm for 60–120 minutes at room temperature.
Note: ProcartaPlex™ assay validation is always performed at a 2-hour incubation at room
temperature. If necessary, the assay can be performed over two days by incubating the 384-well
plate overnight. Shake the 384-well plate for 30 minutes at room temperature at 600 rpm, then
transfer the plate to 4°C and store on a level surface. After overnight incubation, shake the plate for
an additional 30 minutes at room temperature at 600 rpm.
5. Wash the 384-well plate twice following step 3.
6. Add Detection Antibody Mixture and incubate.
a. Add 6.3μL of Detection Antibody Mixture (1X) to each well.
b. Seal the plate with a new Plate Seal, cover the plate with the Black Microplate Lid, and
incubate 30 minutes on a plate shaker at room temperature at 1,400±50rpm.
7. Wash the 384-well plate twice following step 3.
8. Add SAPE and incubate.
a. Add 12.5μL of SAPE solution to each well.
b. Seal the plate with a new Plate Seal, cover the plate with the Black Microplate Lid, and
incubate 30 minutes on a plate shaker at room temperature at 1,400±50rpm.
9. Wash the 384-well plate twice following step 3.
10. Prepare the 384-well plate for immediate or later analysis on a FLEXMAP 3D™ or INTELLIFLEX™.
a. Add 50 μL of Reading Buer into each well.
b. Seal the plate with a new Plate Seal, cover the plate with the Black Microplate Lid, and
incubate 5 minutes on a plate shaker at room temperature at 1,400±50rpm.
c. Remove Plate Seal and run the plate on a FLEXMAP 3D™ or INTELLIFLEX™.
Note: Alternatively, the sealed plate can be stored overnight at 2‑8°C before running on a xMAP™
instrument. Shake the plate at 1,400 rpm for 5 minutes at room temperature, then remove the plate
seal and run the plate.
