Product description
The Invitrogen™ NorthernMax™ 10X Denaturing Gel Buer (Cat. No. AM8676) is a concentrated formaldehyde-based formula for the
preparation of denaturing formaldehyde/MOPS agarose gels. The 10X gel buer is for use with the NorthernMax™ system.
Contents and storage
Contents Amount Storage
NorthernMax™ 10X Denaturing Gel Buer 250 mL 4°C
Prepare a denaturing formaldehyde/MOPS 1% gel
For 100 mL of gel:
1. Melt 1 g agarose in 90 mL nuclease-free water in a microwave or on a hot plate, with frequent agitation, until the agarose is
completely in solution. Transfer the solution to a 50–60°C water bath until equilibrated.
2. Under a ventilation hood, add 10 mL of NorthernMax™ 10X Denaturing Gel Buer to the equilibrated agarose solution and mix well
by swirling.
3. Pour the agarose solution into a gel casting tray in a ventilation hood and allow it to solidify.
The gel should be approximately 0.6 cm thick for ecient transfer.
Once gel has hardened, proceed with electrophoresis in 1X MOPS buer (e.g., prepared from NorthernMax™ 10X Running Buer;
Cat. No. AM8671)
Quality control
•Nonspecific endonuclease activity: A sample is incubated with supercoiled plasmid DNA and analyzed by agarose gel
electrophoresis.
•Exonuclease activity: A sample is incubated with labeled double-stranded DNA, followed by PAGE analysis.
•RNase activity: A sample is incubated with labeled RNA and analyzed by PAGE.
•Physico-chemical testing: pH, conductivity, density, refraction index
