Combs
Single-use combs with 36, 48, 64, or 96 lanes are now available for the 377 DNA
sequencer and the 377 DNA sequencer with XL and 96-lane upgrades.
Advantages of
Single-Use Combs
Single-use combs have two main advantages:
♦Eliminate lane leakage
–These cellulose-based combs swell to fit gel plate irregularities. Plastic facing
lends strength, while the cellulose core expands up to 140% to bridge
variations in the loading region. This lock-in-place mechanism provides a
better seal than existing combs and confines samples within each well.
♦Ease of use
–Wear from reuse (
e.g.
, damaged teeth) is avoided, as these combs are only
used once and discarded. They do not require cleaning or special care.
Recommendations
for Single-Use
Combs
♦Use a standard Mylar (plastic) casting comb. (P/N 401913 or 401915, designed
for the single-use combs, or a casting comb integral to Mylar sharks-tooth combs
may be used.)
♦After gel polymerization, clean and prepare plates and well regions as usual.
♦For 36-lane combs, allow a small quantity of deionized water to remain in the
loading region during comb insertion.
–Insertion may be easier with 48-, 64-, and 96-lane combs if all liquid is
removed.
♦Insert the comb as usual. Ensure that the comb is in uniform contact with the gel
surface.
Note It is expected that the comb will fit loosely when initially inserted.
♦The 36-lane combs (with wetted well region) will swell within about 1 minute and
lock the comb in place.
–For other combs (when inserted dry), add approximately 500 µL deionized
water uniformly across the top of the comb to secure its position.
♦Load samples onto the gel as usual (as with a Mylar sharks-tooth comb).
♦For best results, remove the comb after the samples are initially run into the gel
(2–5 minutes).
–If stagger loading, load the first set of samples, run in, then load the second
set of samples and run for 1–2 minutes. Remove the comb and continue the
run.
Storage of
Single-Use Combs
Store the combs in a desiccated environment.
IMPORTANT Long-term storage in high humidity may cause the combs to swell. Swollen
combs may be difficult or impossible to insert between the plates.
