Product description

Gibco™ GlycanTune™ B+ Total Feed (Cat.No.A29720) is an AGT™ dry‑format, total feeding supplement developed for use in fed‑batch

culture of multiple cell lines. The single‑part supplement can shift glycan proﬁles from mostly G0F to primarily G1F and G2F

glycans. GlycanTune™ B+ Total Feed is an animal‑origin free, chemically‑deﬁned formulation that contains no proteins, hydrolysates,

or components of unknown composition. The product provides an easy‑to‑reconstitute, pH‑neutral solution that can be concentrated from

59.2 g/L (1X; recommended) up to 177.6 g/L (3X), thus allowing delivery of key nutrients in smaller volumes.

Guidelines for maximizing protein glycosylation

To maximize protein glycosylation, GlycanTune™ B+ Total Feed

can be added as a complete standalone feed every 1–3days

or as a continuous feed. To target glycosylation proﬁles, initiate

feeding with EcientFeed™ B+ AGT™ Supplement, then transition

to GlycanTune™ B+ Total Feed. (Both feeds can be added every

1–3 days or as continuous feeds.) The timing of the transition

to GlycanTune™ B+ Total Feed will determine the glycosylation

proﬁle. We recommend testing at least three dierent transition

time points (early, middle, and late culture) to determine the

optimal transition point that will result in the desired glycan proﬁle.

Total feed volume can range from 10–45% (1X) to 3–15% (3X)

of the starting culture volume, depending on the cell line and

process. For best results, we recommend that you initiate feeding

only after reaching mid‑ to late‑exponential phase.

page2.

Reconstitute GlycanTune™ B+ Total Feed

1. Measure 80% of the ﬁnal volume of deionized or distilled

water at room temperature (15°C to 30°C).

2. Add GlycanTune™ B+ Total Feed to the water at 59.2g/L

(recommended), then mix for a minimum of 20 minutes

until completely dissolved. The solution will initially appear

cloudy, but will clear over time.

Note: Depending on your culture needs, GlycanTune™ B+

Total Feed can be concentrated up to 177.6g/L (see

Table1).

3. Using a calibrated vessel, dilute to the ﬁnal volume with

deionized or distilled water. Mix for an additional 10 minutes.

4. Measure pH and osmolality. Conﬁrm a ﬁnal pH of 6.8–7.3

and ﬁnal osmolality of 375–415mOsm/kg.

5. Sterilize immediately by membrane ﬁltration

(positive‑pressure recommended).

Use the ﬁltered solution immediately or store at 2°C to 8°C

protected from light for up to 30days.
