Introduction

Biotin binds with high affinity to avidin, streptavidin and Thermo Scientific NeutrAvidin Protein. The bond formation is

rapid and, once formed, is unaffected by most pH extremes, organic solvents and other denaturing agents. The strength and

specificity of the avidin-biotin interaction makes it useful for developing various assays. Avidin-biotin chemistry is an

effective way to increase sensitivity in several types of immunoassays including ELISA, blotting and immunohistochemical

applications. Most of these assays use conjugates of avidin, streptavidin or NeutrAvidin® Protein with enzymes (such as

horseradish peroxidase or alkaline phosphatase) or fluorophores (FITC). Also, biotinylated secondary antibodies provide

versatility and increased sensitivity via signal amplification.

Procedure for Testing Enzyme Conjugates

A. Additional Materials Required

• Phosphate-buffered saline (PBS; 25mM phosphate, 150mM sodium chloride; pH 7.2; Product No. 28372)

• Horseradish peroxidase (HRP) conjugated to avidin (Product No. 21123), streptavidin (Product No. 21126) or

NeutrAvidin Protein (Product No. 31001)

• Thermo Scientific 1-Step Ultra TMB-ELISA (Product No. 34028)

• Stop solution: 2M H2SO4

B. Method

1. Rinse each well three times with 200µL of PBS.

2. Add 100µL of the HRP conjugate at 0.2-1µg/mL.

3. Incubate the plate at room temperature for 30 minutes.

4. Wash each well three times with 200µL of PBS.

5. Add 100µL the TMB substrate and incubate at room temperature for 10 to 30 minutes.

6. Add 100µL of stop solution. Measure the absorbance at 450nm.
