Installation & Operation
Components
A diffuser (CAT #400-3222-2X) is recommended for
use with the 2x objective (CAT #400-3172-PL) for the
EXC-400 microscope. The diffuser and 2x objective are
sold separately.
The diffuser aides to provide even specimen illumination
across the entire field of view.
The diffuser threads into the bottom of the condenser.
The condenser is provided with the EXC-400 microscope.
Installation & Operation
. Install the 2x objective in the nosepiece, typically in the
open position adjacent to the 4x objective.

Select the 10x objective and perform Köhler illumination as
described in the manual that came with the microscope.
To perform Köhler illumination, the condenser top lens
must be flipped into the light path using the lever on the
left side of the condenser.

Thread the diffuser into the bottom of the condenser.

When using the 2x or 4x objective for observation, flip out
the condenser top lens. Be certain to flip in the top lens
when observing with a 10x or higher magnification
objective.
Option 1: Remove the diffuser when not using the 2x
objective for observation. This option provides the best
illumination and image quality during observation with
other objectives. It also enables the ability to perform
Kohler illumination quickly for optimal contrast and
resolution.
Option 2: Leave the diffuser in place when not using the
2x objective. Adjust the condenser aperture diaphragm
per each objective. This option offers efficiency when
using the microscope. Illumination and image quality are
generally acceptable by most users, despite the diffuser
being installed.
