How to use this manual
This manual along with information contained on product labels and in package inserts should provide
you with all the information you need to operate and maintain the Cytofuge 12 Cytocentrifuge system.
When the information requires special attention, a caution symbol appears next to the italicized
text.
Please pay close attention to the instructions that accompany the notes and symbols as well as the
standard laboratory practices outlined by your facility and local regulatory agencies. The table below lists
all the CAUTIONS and WARNINGS for the Cytofuge
®
12 Cytocentrifuge.
WARNING - Do not expose the cytocentrifuge or its rotor to strong or concentrated acids, bases or
oxidizing agents, aromatic or halogenated hydrocarbons, esters or ketones.
WARNING - Do not use the supplied line cord outside of North America. Use a power cord with a
grounded male connector suitable for the power outlet being used, which is rated at 3.0 Amps or more
and has an IEC320/CEE22 female connector to attach to the cytocentrifuge.
WARNING - Maintain a 300 mm (11.8 inches) clearance envelope around cytocentrifuge during
operation. Do not deposit any potentially hazardous materials within the CLEARANCE ENVELOPE.
WARNING - Inspect instrument for cracks or any physical damage to housing, cover, lid and rotor.
Damage may result in unsafe operation; discontinue use until repairs have been performed.
CAUTION - Do not overfill the concentrators. Addition of excess amounts of liquid to Cell or Filter
Concentrators will result in the fluid being “spun out” during centrifugation.
CAUTION - Do not place the cytocentrifuge on top of other equipment and do not place other objects or
equipment on top of the cytocentrifuge.
CAUTION - The cover interlock by-pass is for emergency use only. Do not attempt to operate the
Cytofuge 12 while the latch release tool is inserted in the housing. Misuse of the interlock bypass may
expose the operator to unsafe conditions.
CAUTION - Before using any cleaning or decontamination methods except those recommended,
operators should check with customer service that the proposed method will not damage the
equipment.
CAUTION - The rotor (CFRT-12) is designed for many years of service but, depending on the
frequency of use, the rotor components may develop cracks over time or if dropped on a hard surface.
Inspect the rotor for damage or cracks before each use. If damage is found discontinue use and
contact customer service or your distributor for a replacement.
CAUTION - Do not lift the rotor (CFRT-12) by the locking button - the lid may release allowing the rotor
bowl to fall. Always lift the rotor by the black center grip.
CAUTION - The rotor (CFRT-12) is intended for use with biological samples only. Do Not place
flammable or reactive substances in the rotor.
CAUTION - While in the resting position, the rotor is designed to maintain the Concentrators at an
angle to prevent fluid from contacting the slide and/or the filter card prior to spinning. Keep the rotor
level during transport to and from the cytocentrifuge to ensure proper concentrator angle.
CAUTION - Do not defeat any safety interlocks while checking the rotor speed. Bypassing interlocks
could result in personal injury.
CAUTION - Unplug the cytocentrifuge from the wall outlet before cleaning or performing maintenance.

CAUTION Do not lean, or place materials, on the cytocentrifuge.
CAUTION - Follow Universal Precautions with all biological specimens, regardless of whether the
specimen is known to contain an infectious agent. Bioseals cannot be relied upon as a sole safeguard
against contamination by pathogens. Always load and unload the rotor under a biological safety hood.

CAUTION The seals in the removable rotor are intended to stop gross fluid leakage only. These
seals should not be relied upon for protection against biohazardous contamination.

Unpacking and Installation
Inspect Packaging
The Cytofuge
®
12 Cytocentrifuge and its accessories are delivered in a single carton. If the cytocentrifuge or
accessories have been damaged in transport, please inform your carrier immediately.
NOTE: Save the shipping carton and packing materials to facilitate return should service be required.

Install
o Place the cytocentrifuge on a solid level surface suitable for operation of laboratory instrumentation.
CAUTION - Do not place the cytocentrifuge on top of other equipment and do not place other
objects or equipment on top of the cytocentrifuge.
o Position the cytocentrifuge out of direct sunlight and away from sources of intense heat or cold.
Improper conditions can cause damage to the housing, rendering unit unsafe to operate.
o See the Appendix for the acceptable range of operating temperature and humidity.
o Remove the foam shipping retainer around the rotor before operating the cytocentrifuge.
WARNING - Maintain a 300 mm (11.8 inches) clearance envelope around cytocentrifuge during
operation. Do not deposit any potentially hazardous materials within the CLEARANCE ENVELOPE.

Connect Power
o Plug the cytocentrifuge into a grounded outlet supplying 100-240 V @ 50-60 Hz.
o Turn on the power switch (located on the rear panel, above the line cord connector).
o When the Cytofuge 12 completes the power-on check it will give a two-beep ready signal. Press the
STOP/OPEN button to release the latch and open cover.
WARNING - Do not use the supplied line cord outside of North America. Use a power cord with a

System Overview
Principle and Intended Use
For in vitro diagnostic use in concentrating cells onto a microscope slide.
The Cytofuge
®
12 Cytocentrifuge is a microprocessor-controlled cell preparation system that uses
centrifugal force to deposit cells onto microscope slides. Samples are centrifuged in reusable or
disposable gasket-sealed chambers (Cell Concentrators), or in disposable chambers (Filter
Concentrators) that include a filter material to absorb and capture suspension fluid during centrifugation.
A dual row 12-position rotor with transparent lid is supplied as standard equipment. The system is
designed to be operated on a laboratory bench or, if potentially biohazardous samples are processed, in
a biological safety cabinet. A cover interlock system prevents operation until the cytocentrifuge cover is
securely latched, and prevents access to spinning parts until the rotor has come to a complete stop.
Speeds from 200 rpm to 2000 rpm and cycle times from 1 minute to 99 minutes may be selected. The
system features three acceleration rates and dynamic braking to protect delicate samples.

CONTROL PANEL
To change the cycle speed use the arrow keys to the right of the SPEED display. Each key press will
change the speed by 10 rpm. Press and hold the key for 2 seconds to change the rpm in 50 rpm
increments.
To change the cycle time use the arrow keys to the right of the TIME display. Each key press will change
the time by 1 minute. Press and hold the key for 2 seconds to change the time in 10 minute increments.
To change the Acceleration rate press the key until the desired rate (high, medium, or low) is illuminated.
To change the Program use the arrow keys to the right of the PROG display.

Error Indicators

12 Cytocentrifuge error conditions are signaled by flashing codes appearing in the TIME
display on the front panel.
To clear an error condition, correct the problem and press the Stop/Open button.

Sample Requirements/Preparation
Overview
The approximate cell concentration of the specimen should be established prior to slide preparation on
the Cytofuge 12. Samples containing higher than optimal cell concentration will produce slides with cells
too closely packed or overlapping. Samples containing too low a cell concentration will yield slides where
cells are difficult to find, count, or examine. The following is a general guideline for sample concentration
based on an average cell diameter of 10 µm:

safeguard against contamination by pathogens. Always load and unload the rotor under a biological
safety hood (See references).

Pre-concentration
For best results, samples low in cellular content should be pre-concentrated. For example, if the original
sample contains about 100 cells/µL, a 10x pre-concentration will provide the 1,000 cells/µL, which is
recommended for the Cytofuge 12.
To pre-concentrate a sample,
1. Transfer 10 mL of sample to a conical polypropylene centrifuge tube.
2. Spin the sample at 1000-1500 xg for 10-15 minutes in a conventional centrifuge.
3. Decant 9 mL of cell-free supernatant.
4. Mix the cell pellet and remaining supernatant by vortexing or agitation of the tube.
5. Pipette appropriate volume of the concentrated sample to Cytofuge concentrators.
Dilution
For best results, dilute samples that have an extremely high cell density. For most applications, buffered
saline or standard tissue culture media (e.g. Geys balanced salt solution) may be used as a diluent.
Adding a drop or two of bovine serum albumin (BSA) promotes cell adhesion to the microscope slide.

Operating Instructions
Opening and Closing the Cover
An electronic operated cover interlock mechanism prevents operation if the cover is not completely
closed and locked, and prevents the cover from opening when the rotor is turning. An operating cycle can
be initiated only if the cover latch is engaged.
Cover Interlock By-pass
In case of power failure or malfunction, the Stop/Open button may not release the cover. In this event
the cover interlock mechanism can be released manually by inserting the manual latch release tool
supplied with the Cytofuge 12 into the small hole located in the front of the housing. Push the tool inward
to release the latch. Remove the tool before re-starting the cytocentrifuge.
CAUTION - The cover interlock by-pass is for emergency use only. Do not attempt to operate the
Cytofuge 12 while the latch release tool is inserted in the housing. Misuse of the interlock bypass
may expose the operator to unsafe conditions.
Using the Removable Rotor
The Cytofuge
®
12 Cytocentrifuge is shipped with the rotor installed and supported by a shipping
retainer. To prevent possible damage to the cytocentrifuge motor mount, it is important to remove the
rotor whenever the cytocentrifuge is moved or shipped to another location.
CAUTION - The rotor (CFRT-12) is intended for use with biological samples only. Do Not place
flammable or reactive substances in the rotor.
CAUTION - The rotor (CFRT-12) is designed for many years of service but, depending on the
frequency of use, the rotor components may develop cracks over time or if dropped on a hard
surface. Inspect the rotor for damage or cracks before each use. If damage is found discontinue
use and contact customer service or your distributor for a replacement.
To remove the rotor:
Open cytocentrifuge cover. Grasp rotor lid from the black
center grip and lift straight up and out of cytocentrifuge.
Never lift the rotor by the center button.
To open the rotor:
With rotor removed from instrument, pull up on the center
button to release lid.
Pull up on the black center grip to remove the lid.
To re-install rotor:
Place lid onto rotor. Press center button to lock lid in place.
Lift rotor by black center grip. Place rotor assembly onto cone located in center of instrument.
CAUTION - Do not lift the rotor (CFRT-12) by the locking button - the lid may release allowing the
rotor bowl to fall. Always lift the rotor by the black center grip.

Spinning Samples
CAUTION Maintain a 300 mm (11.8 inches) clearance envelope around cytocentrifuge during

Loading
1. Press Stop/Open button on front panel to release latch and open cover.
2. Remove rotor from cytocentrifuge and transport to a biological safety hood.
3. Remove the lid and load the concentrators into the rotor (concentrators must be equally
distributed and equally filled to properly balance rotor).
4. Slowly add sample to the bottom of the Filter Concentrator or Cell concentrator well. Avoid
getting droplets onto the walls of the funnel/well.
5. Install lid onto rotor, press center button to lock.
6. Place rotor into cytocentrifuge, assure rotor is fully seated onto cone in center of bowl.
7. Close cover and set spin cycle.

Running a Cycle
1. Use the buttons on the front panel to select the desired acceleration, speed and time for the
cycle. Alternatively, select a program number to retrieve previously saved settings (See
Programming - pg 8). The table below lists the relative centrifugal force (RCF) at several
speeds.
2. Press start to begin cycle. The Cytofuge 12 will spin for the set time and automatically stop.
Note: Settings cannot be changed once the cycle begins.

Cytofuge 12 operating parameters generally depend upon the size and specific gravity of the cells to
be concentrated onto the microscope slide. The operator should experiment with different settings to
achieve optimum performance for specific applications. The following are general guidelines:

Unloading
1. Upon completion of the cycle, the rotor will decelerate to a complete stop, a signal beep will
sound, and the cover latch will automatically release.
2. Lift the cover and carefully remove the rotor and transport to a biological hood.
3. Remove the rotor lid to access the sample concentrators
Programming
The Cytofuge
®
12 Cytocentrifuge can store up to 24 user-defined programs. Each program allows
independent settings for speed, time and acceleration. When a program number is selected, the display
will automatically update to show the speed, time, and acceleration associated with that program.
1. Press the Save button once. The displays will flash indicating that the instrument is in programming
mode.
2. Select the program number to be stored by depressing the up or down arrow keys to the right of the
Program display.
Note: Program 00 contains the default instrument settings and cannot be changed.
Programming can be cancelled at any time by pressing the Stop/Open button. The previous values for
that program number will be retained.
3. Adjust the speed and time values in a similar manner and select the desired acceleration rate.
4. Press the Save button a second time to store the values under the selected program number and exit
programming mode.

Maintenance
Overview
Iris Sample Processing recommends that instrument operators perform periodic inspections and
preventative maintenance on all instruments. Contact customer service or your distributor for optional
repair and calibration services offered for this instrument.
CAUTION - Unplug the cytocentrifuge from the wall outlet before cleaning or performing
maintenance.
WARNING - Inspect instrument for cracks or any physical damage to housing, cover, lid and rotor.
Damage may result in unsafe operation; discontinue use until repairs have been performed.
WARNING - Do not expose the cytocentrifuge or its rotor to strong or concentrated acids, bases or
oxidizing agents, aromatic or halogenated hydrocarbons, esters or ketones.
Cleaning
Clean the outside surfaces and switch overlay panel of the cytocentrifuge with a water-dampened cloth
and mild detergent. Remove rotor from instrument. Clean the inner surface or bowl with a mild detergent,
and if necessary, a disinfectant. Acceptable disinfectants are 70% alcohol or 10% bleach solution.
CAUTION - Before using any cleaning or decontamination methods except those recommended,
operators should check with customer service that the proposed method will not damage the
equipment.
General Cleaning
1. Unplug cytocentrifuge from the electrical outlet.
2. Open the cytocentrifuge cover and remove the rotor.
3. Clean liner and soiled external surfaces with a soft cloth dampened with a mild detergent.
4. Re-install rotor (see Removing and Installing the Rotor).
5. Plug into electrical outlet.
Rotor Disinfection
1. If necessary, remove rotor from the cytocentrifuge.
2. Remove lid from rotor bowl.
3. Remove center seal from rotor bowl.
4. Clean all parts with a soft cloth dampened with a mild detergent.
5. Soak all parts in a 10% bleach solution for 30 minutes.
6. Rinse the parts under running tap water or soak for 5 minutes.
7. Dry the parts with a clean absorbent paper towel or allow to air dry.
8. Replace the rotor seal, re-attach the lid and re-install the rotor in the Cytofuge 12.
Rotor Sterilization
1. If necessary, remove rotor from the cytocentrifuge.
2. Remove lid from rotor bowl.
3. Remove center seal from rotor bowl.
4. Clean all parts with a soft cloth dampened with a mild detergent or if heavily soiled, soak in mild detergent.
5. Place all parts into autoclave at 121 C for 15 minutes.
6. After sterilizing, replace rotor seal, re-attach lid and re-install the rotor in the Cytofuge 12.

Checking/Replacing Rotor Seal
Over time seals may become brittle and develop cracks. To ensure optimal rotor performance, check the
inner and outer seals of the rotor regularly for any tears or cracks. The center seal is user replaceable.
The outer seal is part of the rotor bowl if this seal becomes worn or damaged a new rotor bowl should
be purchased.
Contact customer service or your distributor to purchase a rotor seal kit (P/N 01-006166-001), which
includes the center seal and drive cone o-rings, or a new rotor bowl (P/N 01-006168-001), if necessary.
To replace the center seal, simply pull the seal off of the rotor bowl and press on the seal as shown in the
figure.
Checking/Replacing Drive Cone O-rings
Over time the drive cone o-rings may become either worn or rough. If the o-rings need replacing, remove
the o-rings with a paperclip or tweezers and slide the new o-rings into the grooves. The o-rings are two
different sizes the large o-ring (P/N 23-003019-137) fits the large diameter groove and the small o-ring
(P/N 23-003019-128) fits the smaller diameter groove. Both o-rings and the center seal are included in
the rotor seal kit (P/N 01-006166-001).

Checking the Rotor Speed
Rotor speeds (+5%) may be checked with a stroboscope or photoelectric tachometer.
1. Remove the rotor and locate the reflective patch located on the top of the drive cone.
Close the instrument cover.
2. Set the instrument to the desired speed.
3. Point the tachometer through the transparent cover at the reflective patch.
4. Press the Start button. Allow the instrument sufficient time to reach speed before making
a measurement.
If the instrument fails to maintain set speed within specified tolerance contact customer service
or your distributor.
CAUTION - Do not defeat any safety interlocks while checking the rotor speed. Bypassing
interlocks could result in personal injury.
Troubleshooting
Symptom
Solution
Error indicator flashing, unit will not spin
Refer to Error Code Table of manual to identify and correct problem.
Concentrators are loose and do not fit into rotor
Use only Cytofuge concentrators, other manufactures may look similar but
only Cytofuge concentrators may be used.
No display lights are on and unit will not spin
Main power switch is located on rear panel along with fuse holder. Check
switch position and, if necessary, condition of fuses.
Confirm line cord is plugged into a powered outlet.
Unit vibrates then shuts down
This instrument detected an imbalance and stopped operation. Clear the
error via keypad. Make sure concentrators are balanced within the rotor.
There is a crack in the top cover or housing
Discontinue operation. Contact Customer Service for repair options or a
list of authorized repair facilities.
Cover does not open
Confirm unit has power; press the Stop/Open button.
In the event of a power failure or malfunction, insert latch release tool into
the small hole on front panel. Cover will release and samples can be
removed.

Service
Refer all service to Iris Sample Processing or an Authorized Repair Facility.
Instrument fuse(s) are located on the rear panel. Remove the line cord to access the fuse holder.
Replace with fuses of the same type and rating.

Be sure to complete the Warranty Registration Card as directed.
Decontamination before returning for service:
Any instrument or accessory that has been exposed to blood or other biological materials must be
cleaned prior to shipment to the manufacturer or distributor for service. This decontamination is
required by Federal Law (Title 48 and 49 of the Federal Regulations) and in accordance with the
Environmental Protection Agency’s Regulations for Biohazard Waste Management. Iris Sample
Processing personnel cannot perform this decontamination. Refer to maintenance section for cleaning
and disinfection instructions.
