4. USING Ceveron
®
MFU 500
FIRST-TIME USE
Remove the foam pad which is placed between upper and lower part of the
manifold for protection during shipment.
The power supply is already set to the voltage of your country. The output
voltage is set to 12 Volts and must not be changed.
Checking the desired output voltage
The In-Plug Power Supply as delivered together with Ceveron
MFU 500
is set to an output voltage of 12 V DC.
Check if the DC output voltage is set to 12 Volt
Should the output voltage accidentally be set to a different voltage, it has
to be changed before using it together with Ceveron
MFU 500.
Before changing the output voltage remove the In-Plug Power Supply
from the AC socket
Make sure that the In-Plug Power Supply is not connected to the
Ceveron
MFU 500.
Use an appropriate screwdriver to set the output voltage at the yellow
switch to 12 Volt.
Using higher voltages than 12 Volt will damage the pump inside the
Ceveron
MFU 500
Using lower voltages than 12 Volt will result in poor function of the pump
and of Ceveron
MFU 500.
Change the output voltage only
in a powered-down condition!
Pay attention to the operating
voltage of 12 Volt! Likewise, pay
attention to the firm engagement of the
switch into the respective voltage
position!

OPERATING PROCEDURE
Plug the power supply n in a socket of the regular AC power line and the 12
V power cable into the socket in the grey case
m
.
Take off the upper part of the manifold
b
and place the receiver plate
l
on top of the white spacer block
c
.
Put the manifold collar b in place and put the filter plate
a
into the collar.
Pipette the samples into the filter plate.
Never use the unit
w
ithout
receiver plate! (The filtrate would be
lost and the manifold contaminated.)
Remember the orientation of the
plate, e.g. well A1 on the top left
position.
Make
sure the plate fits well on the
black gasket.
Make sure that the filter plate is in
the same orientation as the receiver
plate! (e.g. well A1 in the top left
position)!
Maximal sample volume for 96
-
well
filter plates: 350 µL
For filtration of plasma we recommend
to use 200 µL/well
Note: It is not necessary to fill all wells
of a plate.
9
10
Seal the filter plate tightly with a plate sealer foil (provided).
Stop Cock
k
has to be in the open position (in line with the tubing
i
).
Press the start button e.
Press the filter plate
a
Note: If a turbid plasma sample
cannot be filtered completely
within the preset time, repeating the filtration will not have an effect as
the filter is likely blocked! See 8. Trouble Shooting
Release the vacuum
After complete ventilation of the unit, first take off the filter plate
a
and then
the manifold collar b.
Finally remove the receiver plate
l
containing the filtrates
To obtain the best possible
vacuum, the whole plate has to
be sealed independent off the
number of filled wells. Make sure
the foil is tightly put on the surface
of the filter plate. We recommend
to use a fresh foil for every
filtration.
After the pump starts,
press the
filter plate
a
aa
a
shortly but tightly to
the manifold collar (b to ensure
tight fitting of the plate. The pump
stops automatically after ~ 3
minutes, the time required for
filtration of 200µL plasma.
To release the vacuum,
press the
ventilation valve button f
ff
f. The
pressure will fall to 0 at the
pressure gauge
h

5. APPLICATION NOTES
Generation of micro particle free plasma samples and recovery of micro
particles from platelet poor plasma:
Ceveron
®
MFU 500 is designed to be used for filtration of fresh, clear,
platelet poor plasma samples.
For turbid plasma samples pre-filtration through another filter with larger
pores is required.
What is the efficiency for removal of micro particles ?
After filtration of plasma spiked with 10,000 MP/µl only 0.4% ± 0.004% of
MPs were found in the filtrate.
Recovery of micro particles from the filter plate
MPs can be recovered from the filter plate by rinsing each well with 200µL
of PBS or micro particle free plasma. Rinsing is performed by gentle
pipetting the sample for 3 to 5 times and then pipetting off the sample.
Recovery rates are in general > 85 %

6. REPLACING O-RING AND GASKET
An accessory kit containing an additional O-ring, gasket and Allen wrench is
included with each unit of Ceveron
®
MFU 500. O-ring and gasket should be
replaced when worn or vacuum leakage is observed.
For replacing the black gasket, the 12 screws have to be loosened by using
the Allen wrench included in the accessory kit. The O-ring can simply be
replaced manually.
7. CLEANING AND STORAGE
Basic preventive maintenance requires regularly cleaning of Ceveron
®
MFU
500.
Use a mild soap, standard laboratory detergent, a dilute solution of sodium
hypochlorite, or ethyl alcohol to clean all surfaces of the manifold. After
cleaning, rinse off any residual liquid with a soft cloth or paper towel
dampened in clean water, then wipe dry.
Should there be any liquid leakage into the manifold base or the vacuum
tubing, it can be removed by rinsing the base with distilled water while the
vacuum pump is running (the pump is water resistant). After rinsing with
water, air should be pumped through the unit at least for 2 runs to dry out
the system
Other than occasional cleaning, the Power Supply is maintenance-free. For
cleaning the case, use a clean, lint-free, antistatic and dry cleaning cloth.
Store in a controlled environment at a temperature between 15°C and 30°C.
